Purge the optical system with nitrogen
for 5 min before turning spectropolarimeter on.
The nitrogen flow rate is set to 15
liters/min. Do not adjust the regulator settings.
Turning ON the “Power” switch on the
instrument front panel does not turn on the light source. The light
source is lit automatically when the measurement program is started.
If the instrument is idle during the
course of an experiment, keep the light source lit. Turning the light
source ON/OFF frequently shortens its lifetime.
If the slit width is set manually, do not
widen the slit more than required. This will slow down the
deterioration of the detector.
Do not set the HT voltage below 170
volts
Shut off the Isotemp water circulator
whenever the light source is turned OFF otherwise water will condense
on the light source unit, deteriorating its optical components.
Instrument Start-Up
Open the nitrogen cylinder and allow gas to purge the optical
system for 5 minutes.
Turn on the spectropolarimeter and the PC
Open the “Spectra Manager” software
Select “Spectrum Measurement” program to initialize the
polarimeter and light the lamp (this will take 1-2 min)
Note: When the light source is lit, the “Lamp” LED of the
Amplifier panel (on front of instrument) will be green.
Obtaining CD Spectra
Open the Spectrum Measurement program
Select the “Measurement” menu, then “Parameter”
Place the 10mm cell in the sample holder
Click “Start” from the “Measurement” menu to begin data
acquisition
After acquiring the measurement, the Spectra Analysis program
opens automatically
Save the spectrum by using the “Save As…” function from the “File”
menu
If you want to use the temperature
control...
In the Spectrum Measurement program, select “Measurement” →
“Accessory”
Make sure “JASCO Peltier Type (Single, RTE on)” is selected under
“Temperature”
To set the desired temperature, click “Control” → “Accessory”
Input the temperature and hit “Apply”
To turn the lamp ON/OFF:
In Spectra Manager choose “Instruments” → “Start” or “Stop”
