A Nuclear Receptor FXR Responsible Element in bile salt export pump promoter
Bile salt export pump (BSEP) is responsible for bile acid removal from the liver and its expression is transcriptionally regulated by nuclear receptor farnesoid-X-receptor (FXR). Impairment of BSEP expression or function is associated with a variety of cholestastic liver injuries. FXR-promoted transactivation of BSEP is mediated through FXR-responsible element (FXRE) in the BSEP promoter. We have identified a potential novel FXRE in the BSEP promoter. The objective of this project is to molecularly and functionally characterize the putative FXRE using regular molecular cloning, cell culture, transfection and promoter reporter assays.
